A Study on Histological Changes in the Gonads, and Changes of water and Electrolyte Contents in Ovary, Uterus, and Adrenal gland of Immture Rabbits by Chlorpromazine Administration


Abstract
Chlorpromazine is used, as a tranquilizer, not only to the patients of toxemia of pregnancy but also to some other conditions in the Obstetric and Gynecologic field. Although chlorpromazine has prominent effect as tranquilizer, it has a number of untoward side effects too.
An attempt was made to clarify the effect of Chlorpromazine to the gonads and changes of water and electrolyte contents in the uterine and adrenal tissue of immature female rabbits weighing about 1.5kg. A group of rabbits preconditioned with 50 I. U. /kg of HCG and another group with 50mg/kg of chlorpromazine plus 50 I. U. /kg of HCG were observed in 24 hours after preconditioning them.
Water content was determined by drying method in 100¢ªC oven until the weight remained constant. Flame-photometry was utilized for the determination of Na and K levels, Schales and Schales method for Cl content and Kovac and Tarrnoky¢¥s method for Ca and Mg contents. The Results obtained are summarized as follows;
1. The HCG preconditioned rabbits showed acceleration in maturation of the folliclels and ovulation in the ovaries, while the chlorpromazine plus HCG preconditioned group showed no suppression in ovulation.
2. Changes in electrolyte levels in serum of the HCG preconditioned rabbits showed an increase in K and Mg levels and a decrease in Cl, Na and Ca levels. In the chlorpromazine plus HCG preconditioned group, on the contrary, K and Mg were decreased in each level and Ca was increased in its level.
3. Contents of water and electrolytes in the uterine tissue of the HCG precoditioned group showed an increase in water and Cl levels and a decrease in Na, K and Mg levels. The chlorpromazine plus HCQ preconditioned group showed a decrease in water, Na, Cl and K levels and a marked decrease in Mg level of uterine tissue.
4. The HCG preconditioned group showed an increase in water, Ca and Mg levels and a decrease in Na level of the adrenal tissue. water and all electrolytes in the adrenal tissue of the chlorpromazine plus HCG preconditioned group was decreased than those of the HCG preconditioned group.
5. Water content in the adrenal tissue was higher than that of the uterine tissue in the normal group, HCG preconditioned group and chlorpromazine plus HCG preconditioned one.
6. Cl content in the adrenal tissue of the normal group and HCG preconditioned one showed higher level than that of the uterine tissue, on the other hand, no significant difference in Cl content was noted between both the adrenal and the uterine tissue in chlorpromazine plus HCG preconditioned one.
7. Na content in the adrenal tissue was higher than that of the uterine tissue of the normal group, HCG preconditioned group and chlorpromazine plus HCG preconditioned one.
8. K contents of both the adrenal and the overy of normal group and chlorpromazine plus HCG . preconditioned one showed very similar to each other in their levels. on the other hand, K content in the adrenal tissue of HCG preconditioned one, was increased.
9. Ca content in the adrenal tissue was remarkably higher than that of the uterine tissue in the normal group, HCG preconditioned group and chlorpromazine plus HCG preconditioned group and chlorpromazine plus HCG preconditioned one.
10. Mg content in the adrenal tissue was higher than that of the uterine tissue in the normal group, HCG preconditioned group and chlorpromazine plus HCG preconditioned one.
